The Effect of Sosokmyung-tang extract on Hypercholesteremia in vivo Cytotoxicity in PC-12 cells induced by amyloid ¥â-protein No Production in RAW cells stimulated by lipopolysaccharide in vitro



Abstract
The effects of Sosokmyung-tang on hypercholesteremia, cytotoxicity in PC-12 cells induced by amyloid ¥â-protein (25-35) and NO production in RAW cells stimulated by lipopolysaccharide were evaluated in this study.
This study was performed to investigate that Sosokmyung-tang would be useful for cerebrovascular diseases and dementia.
In the case of hypercholesteremia, Sprague-Dawley rats were used and divided into three group at random. Normal group was given a general diet, control group was given 1 % cholesterol diet and normal saline for 14days, and experimental group was given 1 % cholesterol diet and 157.2§·/250g/day of Sosokmyung-tang extract for 14days. The multiple parameter of hypercholesteremia included total cholesterol - triglyceride HDL-cholesterol . LDL-cholesterol . VLDL-cholesterol in the serum.
In the case of cytotoxicity in PC-12 cells induced by amyloid ¥â-protein (25-35), we investigated the cytotoxicity of Sosokmyung-tang extract when added it to PC-12 cells and the protective effect on cytotoxicity when added it to PC-12 cells induced by amyloid ¥â-protein (25-35).
In the case of NO production in RAW cells stimulated by lipopolysaccharide, we investigated the effect on NO production when added Sosokmyung-tang extract to RAW cells stimulated by lipopolysaccharide.
The results were obtained as follows:
1. Sosokmyung-tang significantly decreased total cholesterol, triglyceride and LDL-c-holesterol whereas there wasn¢¥t shown any significance in HDL-cholesterol and VLDL -cholesterol.
2. Sosokmyung-tang didn¢¥t shown the protective effect on cytotoxicity in PC-12 cells induced by amyloid ¥â-protein (25-35).
3. Sosokmyung-tang significantly decreased NO production in RAW cells stimulated by lipopolysaccharide.
From the above results, it was concluded that Sosokmyung-tang can be effectively applied to cerebrovascular diseases but can¢¥t to dementia.
